Ethics approval and consent to participate
The study was approved from research and ethical committee, International Islamic University, Islamabad, Pakistan, under the case No. IIU (BI & BT) FBAS-2017 and all the experiments were performed after taking the written informed consent of the subjected family and written consents were obtained from the parents or legal guardians of participant under the age of 16.
Consent for publication
We obtained written informed consent for publication of identifying images or other personal or clinical details was obtained from all of the participants. In the case of minors (Individuals younger than the age of 18) consent for publication were obtained from their parents or legal guardians.

The 3D protein structure model was built by using SWISS-MODEL. Winccot software was used for introducing mutations to structure and Structural effects of a mutation are analyzed through HOPE server.
